If you have an early version of the DLL, it may be that PEDRV was introduced later - or it may be your problem. Is the old machine still around so you can see if it has PEDRV.SYS? Does your software give any error messages? Is this the same PC where it used to work under NT? If not, have you tried any of the P&E software? In order to work, the DLL needs to use several drivers: VICHW11.SYS, PEDRV.SYS (and PEUSBV1.DLL and PEUSBV1.SYS for the USB pod). These are SUPPOSED to work without being pre-installed (the dll loads and unloads them), but I believe that will only work if you have administrator privileges on your PC. Otherwise, you may need to have your administrator run the .reg files that P&E ships with the dll (VICHW11.REG and PEDRV.REG) to install the drivers.
